At this moment, each time I want to synchronize a folder, I need to select the local folder to synchronize to.

It would be great if I could set up one or more "Sync Folders" that I can synchronize with the click of a button.

You can just leave the synchronization configuration in the Transfers window and restart the transfer to synchronize again.
